摘 要:目的 建立测定富马酸喹硫平血药浓度的方法。方法 应用反相高效液相色谱技术,YWG-C_(18)(150mm×4.6mm,10μm)色谱柱,以氯氮为内标,乙醚为萃取剂,甲醇-水-四甲基乙二胺(63:45:0.3)为流动相,冰乙酸调节pH至5.80,流速1mL·min^(-1),检测波长250nm,柱温40℃。结果 富马酸喹硫平及内标与血清杂质可完全分离,保留时间分别为4.6min和6.1min,在10~2400ng·mL^(-1)浓度范围内呈良好的线性关系(r=0.9998),平均回收率98.2%~101.1%,日内、日间RSD在2.7%~6.9%之间,最低检测浓度为5ng·mL^(-1)。结论 该方法操作简便,结果准确,专属性强,具有一定的实用价值。Objective To establish a RP-HPLC method for determination of quetiapine fumarate serum concentration. Methods Instruments, reagents, and conditions adopted in the analysis were: A YWG - C18 column (150mm×4.6 mm,10μm), chlordiazepoxide was used as internal standard, the detective wavelength was 250nm,the mobile phase of MeOH: H2O: TEMDA =63: 45: 0.3 ,its pH was adjusted to 5. 80 with glacial acetic acid, the flow rate was 1.0 mL·min-1 and the column temperature was 40℃. Results Quetiapine fumarate and internal standard can be completely separated with a liner range of 10 - 2400ng · mL-1 ,and correlation coefficients was 0. 9998. The lowest detective concentration was 5 ng · mL-1. The average recovery was 98. 2% - 101. 1% with RSD of intra - day and inter - day assay were within 2.7% -6.9%. Conclusion A simple , accurate, selective and suitable method for determination of quetiapine fumarate serum concentration was established.
