摘  要:目的 探讨碱性成纤维细胞生长因子(bFGF)和重组人骨形成蛋白(rhBMP)分别及联合作用对人牙周膜细胞(PDLCs)增殖的影响;优选其最佳显效浓度。方法 选取因正畸治疗需要拔除的第一前磨牙,刮取根中1/3牙周膜组织作为标本,采用牙周膜细胞的体外培养技术,用四唑盐比色法(MTT)和酶动力学方法,动态观察bFGF、rhBMP梯度含量分别及联合作用对人PDLCs增殖的影响。结果 bFGF、rhBMP分别作用均能促进PDLCs的增殖;bFGF(10)+rhBMP(200)(最佳浓度之和)促PDLCs增殖作用均较其分别作用更加显著;动态观察bFGF(10)+rhBMP(200)作用于PDLCs1~7d,显示出连续递增的促增殖趋势。结论 bFGF、rhBMP分别作用均能促进人PDLCs的增殖,联合应用则更具有协同作用;其中bFGF(10)+rhBMP(200)Objective To find out the effects of basic fibroblast growth factor(bFGF) and recombinant human bone morphogenetic protein (rhBMP)on the proliferation of human periodontal ligament cells (PDLCs) respectively or conjointly and to select their optimal density. Methods The first premolars extracted due to orthognathous treatment were selected and the one - third PDL tissues of fang' s intermediate were scraped as samples PDLCs culture technique, MTT assay and enzyme kinetics methods were used to observe dynamically the effects of gradient contents of bFGF and rhBMP on the proliferation of PDLCs respectively or conjointly. Results Either bFGF or rhBMP can promote the proliferation of human PDLCs;significantly greater proliferation was found in bFGF(10) + rhBMP(200) (sum of the optimal density) compared with that of each;bFGF(10) + rhBMP(200) dynamically acted on PDLCs from 1 to 7 days and indicated continuously promoting proliferation trend. Conclusion Either bFGF or rhBMP can promote the proliferation of human PDLCs, and combined two actions had synergistic function; bFGF(10) + rhBMP(200) was optimal density.
