摘 要:目的:在基因组水平上探讨乌三颗粒抗肿瘤的分子生物学机制。方法:抽提经乌三颗粒治疗的Lewis肺癌组织和未经治疗的对照组瘤组织总RNA并纯化mRNA,分别逆转录并进行荧光标记,制作成cD-NA探针,与小鼠MGEC-20S型基因芯片杂交,筛选出两组间差异表达基因。结果:两组间存在差异性表达基因45条,与对照组比较,中药组与恶性肿瘤浸润、转移相关的基因表达下调,与促进细胞凋亡的相关基因表达上调。结论:基因芯片技术能高通量分析中药乌三颗粒防治Lewis肺癌的相关靶基因,为进一步揭示中药抗肿瘤机制提供了新的思路和前沿性生物技术手段。Objective: To investigate the molecular biological mechanism, on genomic level, of using Wusan Granule in treating Lewis lung carcinoma. Methods: The total RNAs were isolated from carcinoma tissue of two groups, treated or un-treated by Wusan Granule. They were purified, both mRNA were reversely transcribed to the cDNAs and labelled with fluorescence to prepare the cDNA probe, which was hybridized with mouse MGEC-20S gene chip to screen the different expressed genes between the two groups. Results; There were 45 different expressed genes between the two groups. As compared with the group un-treated with Wusan Granule, in the treated group, the tumor infiltration and metastasis related gene expressions were down-regulated and the tumor cell apoptosis related gene expression up-regulated. Conclusion: The cDNA microarray technique is a high-throughout approach to screen the Wusan Granule anti-tumor related target genes, which provides new pathway and advanced biological means for studying of the scientific entity of Chinese herbal medicine in treating tumor.
