细胞增殖周期调控因子P16和CyclinD1在食管癌中的表达和意义

摘　　要：目的　探讨细胞增殖周期调控因子 p16和 cyclin D1在食管癌中的表达和意义。方法　应用免疫组织化学 ABC法检测 6 2例食管癌中 p16、cyclin D1的表达。结果　在食管癌 p16蛋白表达阳性率为 4 8.39% (30 / 6 2 ) ,明显低于癌旁正常粘膜 76 % (38/ 5 0 ) ,染色强度大部分为弱阳性、阳性 ;cyclin D1蛋白表达阳性率为 6 9.35 % (4 3/6 2 ) ,明显高于癌旁正常粘膜 2 8% (14 / 5 0 ) ,染色强度大部分为阳性、强阳性。两者比较有显著差异性 (P<0 .0 5 )。前者的表达与淋巴结转移无关 ,而后者的表达与淋巴结转移有关。结论　食管癌的分子发病机制可能涉及 CDK4 /细胞周期素 D1和Objective To study the expression and significance of cell division cycle regulators p16 and cyclinD1 in human esophageal cancers.Methods The expression of p16 and cyclinD1 in 62 cases of human esophageal cancers was studied with ABC immunohistochemical technique.Results The positive rate of p16 protein in esophageal cancer was 48 39%(30/62),much lower than 76%(38/50)in normal mucosa,the intensity of staining of most cases was weak,but in some of them was strong.The positive rate of cyclinD1 protein was 69 35%(43/62),much higher than 28%(14/50)in normal mucosa,the intensity of staining of most cases was strong and in some very strong.Two fairly have notable difference( P <0 05).The positive rate of p16 protein did not correlate with lymph node metastasis of the tumor,The positive rate of cyclinD1 was correlated with lymph node metastasis of the tumor.Conclusion The abnormality in the negative feed back regulatory pathway of cyclinD1/CDK4,and p16 may be involved in the molecular mechanism of esophageal cancer development.
