摘  要:目的建立人脊索瘤细胞系,为脊索瘤研究提供实验模型。方法取经病理证实的新鲜脊索瘤手术标本(取自于1例42岁男患者行手术切除后的肿瘤),进行体外原代组织块培养。对存活细胞进行形态学观察、组织化学染色、细胞周期检查、染色体分析、电镜观察、异种移植和体外侵袭实验等。结果建成细胞系CM-319,经近两年的体外培养,已连续传代百余次。其形态学表现、组织化学染色、电镜观察和异种移植等均符合脊索瘤细胞特征。细胞倍增时间为33h,细胞周期测定显示:G1期为55.6%,G2期为21.9%,S期为22.5%,G2/G1为1.90。染色体具有亚三倍体核型。异种移植成瘤率100%,具有侵袭性。结论CM-319是一株人脊索瘤细胞系,可用于对脊索瘤的研究。Aim To establish a human chordoma cell line for use in the chordma research.Methods The specimen which was identified pathologically as chordoma were cultured,using primary tissue culture in vitro.The surviving cells were analyzed by morphology, histochemistry,cell cycling analysis, karyotype analysis,electron microscopic observation,heterotransplantation and study of invasive capacity in vitro.Results The newly established cell line which was designated CM 319 has been maintained in continuous culture for over 100 generations in two years.Morphological observation, histochemistry aspect, electron microscopic observation and heterotransplantation showed that it had the common features of chordoma. The doubling time of cells was about 33 hours.The cell cycle analysis showed:G1 55.6% ,G2 21.9% and S 22.5% ,G2/G1=1.90.The chromosome analysis showed a hypotriploid feature and success rate of heterotransplantation was 100% . It has capacity of invasion in vitro.Conclusion The CM 319 appears to be a cell line derived from human chordoma cells and might serve as a model for further studies of chordoma. <P>
