摘 要:目的 :应用大鼠短期实验模型进行Ge- 132对黄曲霉毒素B1(AFB1)诱癌的化学预防研究。方法 :实验大鼠按不同处理方法分为 5组 :A组高剂量Ge - 132 ;B组 :低剂量Ge - 132 ;C组 :给AFB1前高剂量Ge -132 ;D组 :给AFB1后高剂量Ge- 132 ;E组 :正常对照。在第九周动物处死后肝组织作低温石蜡连续切片 ,分别染γ -谷氨酰转移酶 (γ -GT) ,血清和肝组织测定其超氧化物歧化酶 (SOD)活性。结果 :Ge - 132具有抵抗AFB1降低SOD活力的作用 ,Ge - 132的摄入时间、剂量可影响大鼠体内SOD活力和γ -GT灶的大小。结论 :实验结果提示Ge - 132的抗氧化作用与抑癌作用有关 ,是一种安全、有效的肝癌化学预防剂。Objective:To determine the protective effect of Carboxyethylgermanium (Ge-132)during AFB1-induced hepato-carcinogenesis for Wistar short term model experiments.Methods:The animals were divided into 5 groups: A: high dosage of Ge-132;B:lower dosage of Ge-132; C: high dosage of Ge-132 before AFB1 exposure; D: high dosage of Ge-132 after AFB1 exposure;E: normal control group.All of the animals were killed at the end of the eight-week experiment .The slides of the liver tissues were stained for γ-Glutamyltransferase (γ-GT) . The activity of SOD in serum and in the liver tissues was measured.Results:Ge-132 possessed the remarkable resistance effects on AFB1 reducing Superoxide Dismutase(SOD) activity in vivo during AFB1-induced hepato-carcinogenesis for short term model experiments. The SOD activity and the size of r-GT can be effected by the time and the dosage of Ge-132 intake.Conclusion:The results indicate that antioxidative ability of Ge-132 is relate to its anticarcinogenicity. Ge-132 can be used as a safe and effective chemoprophylaxis agent.
