摘  要:目的观察参与细胞发育分化和细胞非对称性分裂的notch信号传导路的分子Numb在体外培养的神经干细胞内的分布和表达,以期为在体外控制神经干细胞的分化提供新的线索。方法取孕14d(E14)Balb/c近交系小鼠胚胎,采用无血清培养技术,在体外进行神经干细胞克隆的培养传代,经过免疫细胞化学鉴定后,用Numb特异性引物对培养的神经干细胞进行RT-PCR分析,PCR产物经克隆测序后,用地高辛标记,对神经干细胞克隆进行原位杂交。结果在培养的神经干细胞内有大量细胞(>95%)均为Numb基因表达信号阳性;干细胞克隆内部中心和边缘相互间未见明显差别。结论Numb基因可能在神经干细胞的增殖和分化过程中起重要作用。Aim To determine the expression and distribution of morphogen molecule- Numb participating in notch signal conduction in asymmetric cell division in cultured neural stem cells in vitro and provide new clues for differentiation controlling of neural stem cells in vitro.Methods Pregancy 14 d(E14) Balb/c inbred mice were used.Cultured neural stem cell clones which were isolated and expanded in serum free medium in vitro were subjected to RT PCR after being transferred several times in vitro and identified by immunochemistry.The amplification products was sequenced and labeled by digoxingemin and in situ hybridization technique was used to detect the cryosections of the neural stem cell clone.Results Large amount of cells(>95% ) in the stem cell clone could be detected positive for signal of Numb gene, there was no significant difference between the positive cells and the center and peripheral of the stem cell clone.Conclusion Morphogen molecule Numb may play an important role in the proliferation and differentiation of neural stem cell.
