摘 要:目的观察胶质瘤C6细胞伽玛刀治疗后基因蛋白水平表达的变化。方法胶质瘤C6细胞在培养瓶中培养,接受不同剂量(5.0 Gy和6.22 Gy)的伽玛刀照射,应用免疫组化技术和图像分析研究p16基因和C-myc基因的表达。结果伽玛刀治疗显著地抑制了C-myc基因蛋白水平的表达,激活了p16基因蛋白水平的表达。实验组C6细胞C-myc基因灰度值为154.4±4.7(5.0 Gy),161.7±5.8(6.22 Gy);对照组为130.28±2.4。实验组C6细胞p16基因灰度值为155.4±2.0(5.0 Gy),124.9±7.1(6.22 Gy);对照组为167.1±6.2。结论胶质瘤C6细胞的凋亡与激活了p16基因蛋白水平的表达和抑制了C-myc基因蛋白水平的表达相关。Objective To observe the change in gene protein expression after the treatment by gamma knife in glioma cells. Methods Glioma C6 cells proliferated in vitro were treated by gamma knife with dose of 5.0 and 6.22 Gy.The pl6 gene and C-myc gene expressions were determined by immunohislochemical technique and image analysis.Results The C-myc gene protein expression in glioma C6 cells was significantly decreased by gamma knife (P<0.01),and the grey value in control group was 130.28±2.4 and in gamma knife treatment groups was 154.4±4.7 (5.0 Gy) and 161.7±5.8 (6.22 Gy).The pl6 gene protein expression in glioma C6 cells is significantly increased by gamma knife (P<0.01),and the grey value in control group was 167.1±6.2 and in gamma knife treatment groups was 155.4±2.0 (5.0 Gy)and 124.9±7.1 (6.22 Gy).Conclusions One of the mechanisms of gamma knife inducing glioma C6 cell apoptosis is correlated with the increase in p16 gene expression and decrease in C-myc gene expression.
