摘  要:目的:探讨心脏营养素-1(CF-1)及其受体亚基gp130,白血病抑制因子受体-β(LIFR-β)在失神经骨骼肌中的表达规律。方法:选用11组Swiss小鼠,每组10只,切断其右侧胫神经,分别于不同时间点取失神经支配的腓肠肌,用Northern blot法测定肌肉中CT-1和LIFR-β,gp130的mRNA含量,探讨三者的表达规律。结果:正常肌肉组织中CT-1的表达量较高,而2个受体亚基只有低水平表达;随着骨骼肌失神经支配时间的延长,肌肉细胞中CT-1的表达进行性下降:LIFR-β和gp130均为先增高后降低,但前者的表达高峰出现在失神经支配后24h,而后者则在神经切断后1wk表达最高。结论:骨骼肌细胞内源性CT-1表达不足,对肌肉的保护作用减弱可能是失神经肌肉早期萎缩的一个重要因素。受体gp130/LIFR-β的表达增高为进一步研究应用外源性CF-1来防治失神经肌肉萎缩提供了理论依据。AIM: To investigate the expression pattern of cardiotrophin-1 (CT-1) and its receptor in denervated skeletal muscles. METHODS: The tibial nerves of the mice in the 11 groups were dissected and the denervated skeletal gastrocnemius muscles were got to assay the mRNA of CT-1 and its receptor using Northern blot analysis. RESULTS: CT-1 and its receptor mRNA were all expressed in normal muscle and showed their expression pattern respectively after denervation. The expression of CT-1 declined continuously. LTFR-β and gp130 (the two subunits of the receptor) both augmented followed by gradual descent. The difference was that the expression peak of the former appeared at 24 h after devervation, while that of the latter was at one week after the operation. CONCLUSION: The inefficient expression of intrinsic CT-1 in muscle cells, and its diminished protective effect to the muscle is one of the important factors of the early atrophy after devervation. The augmented expression of receptor is significant for the study of the effect of exogenous CT-1 on the prevention and therapy of denervated atrophy.
