摘 要:目的 用3H-脱氧葡萄糖(3H-DG)掺入判断6MV X线照射A549肺腺癌多细胞肿瘤球体(MTS)的疗效。方法 MTS分别作低剂量分割和单次照射,中等剂量单次照射。MTS消化成单个细胞后用流式细胞仪检测细胞周期和凋亡比率。96孔盘每孔1个MTS,加37 kBq 3H-DG后掺入60 min,吸出MTS抽滤后测量β计数。结果 2~8 Gy低剂量照射后未见明显细胞周期阻滞和凋亡,分割照射组MTS的摄取减低,4.84 Gy单次照射后的摄取明显高于其他各组(P<0.05);15~30 Gy照射后,细胞G2-M期阻滞百分率增加,各照射组3H-DG摄取均小于对照组(P<0.05),随照射剂量递增,MTS摄取3H-DG的能力降低;15 Gy照射后第10天有活细胞增殖,25或30 Gy照射后第40天有活细胞增殖。结论 用3H-DG可以监测高能X线照射A549肺腺癌MTS的疗效,不同剂量照射后球体细胞的再增殖时间对于设计放疗方案和安排18F-FDG显像时间有指导作用。Purpose: To investigate the treatment effect of 6MV X - ray irradiation on A549 lung adenocarcinoma multicellular tumor spheroids (MTS) using 3H-Deoxyglucose(3H-DG). Methods: Low doses of 2,4,6,8 Gy were used as fractional radiation, with 2 Gy per times and every 24h. Single radiation were done with the following doses, they were 4.84 Gy which was equal to 6 Gy of fractional radiation, 7.31 Gy equal to 8 Gy of fractional radiation, so did 15,20,25,30 Gy. After digested the MTS into single cells, the ratios of the cell cycle and the apoptosis were scanned by flow cytometry. Every MTS was placed into every hole of 96 holes plate, after 37kBq of 3H-DG was mixed into every hole for 60 min, the MTS were aspirated, filtered and then measured the β counts. Results: After irradiated with 2-8 Gy, there were not remar-kable changes in the cell cycle and apoptosis. The uptake of 3H-DG in the MTS after fractional irradiation with 4, 6,8 Gy were lower than that of 2 Gy, whereas in the single irradiation groups with 4.84 and 7.31 Gy, the uptakes were higher than other groups, among them the counts of the 4.84 Gy group were the biggest (P 3H-DG decreased accordingly (P 3H-DG. The different re-growth time for different doses could have reference value in design the radiotherapy plan or arrange the 18F-FDG examination.
