摘  要:目的:研究丹参酸乙(SA-B)对大鼠肝星状细胞细胞内氧化水平及增殖细胞核抗原(PCNA)的影响。方法:原位灌注法分离正常大鼠肝星状细胞,丹参酸乙直接添加于原代培养的肝星状细胞;2`7`-二氟二氢荧光素(DCFH)掺入反映细胞内氧化程度,Western blot蛋白印迹法检测PCNA含量。结果:1μm和10μm丹参酸乙可减低活化的以及经丙二醛(MDA)刺激后的肝星状细胞的细胞内氧化程度。1μm和10μm丹参酸乙都可抑制活化的以及经MDA刺激后的肝星状细胞的PCNA表达量。结论:丹参酸乙可降低细胞内氧化程度,抑制MDA刺激后肝星状细胞PCNA的表达,丹参酸乙的抗氧化和抗增殖这两方面的作用可能有一定的联系。Objective: To investigate the effects of Saltanic-acid (SA) B for cellular oxidation and PCNA of hepatic stellate cells in rat. Methods: Hepatic stellate cells were derived from normal rat by perfusion and digestion in situ and density gradient centrifugation with Nycodenz. Then cells were treated by various concentrations of drugs. The oxidation of cell were reflected by DCFH incorporation; the content of PCNA were tested by Western bolt. Results: Both 1μm and 10μm SA-B decreased the cellular oxidation, the result was same for MDA-stimulated cells. Both 1μm and 10μm SA-B inhibited the expression of PCNA and also inhibited the expression of PCNA on MDA-stimulated hepatic stellate cells. Conclusion: SA-B could inhibit the cellular oxidation and the expression of PCNA. We can conclude that there are some relations between anti-oxidation and anti-proliferation of Salianic-caid B.
