摘  要:[目的]研究结肠癌瘤细胞对自体肿瘤浸润淋巴细胞(TIL)、肿瘤引流淋巴结的淋巴细胞(LNL)、外周血淋巴细胞(PBL)的影响。[方法]比较结肠癌患者自体肿瘤细胞对体外培养TIL、PBL、LNL细胞增殖、诱生细胞因子IL鄄2、TNF鄄α、IFN鄄γ水平的影响。[结果]结肠癌患者的TIL和PBL、LNL相比,CD8+细胞增加,CD4+细胞数减少。PBL和LNL对PHA、rhIL鄄2刺激产生的细胞增殖能力比TIL强。PBL能产生较高水平的TNF鄄α和IFN鄄γ,而IL鄄2产生水平较低;LNL产生高水平的TNF鄄α和IL鄄2,但仅产生低水平的IFN鄄γ。TIL产生的TNF鄄α、IFN鄄γ水平较PBL、LNL低。[结论]结肠癌患者自体肿瘤细胞对不同来源淋巴细胞的细胞因子分泌有不同影响,PBL可能是结肠癌过继性细胞免疫治疗较为理想的CTL前体细胞来源。To study the effect of autologou s tumor cells from colorectal cancer patients on the im-munologic function of T umor-Infiltrating Lymphocytes(TIL),Lymph Node Lymphocytes(LNL)and Peripheral Bl ood Lymphocytes(PBL).Tumor samples and TIL,LNL,PBL were obtained from18 patients with Colorectal Carcinoma.The effect of autologous tumor cells on TIL,L NL and PBL in patients with colorectal carcinoma,including cellular phenotype,th e proliferation response to mitogens and the capability of prod-ucts(IL-2,IFN-γ,et al.)was compared.TIL showed an enrichment of CD 8+ cells with a correspond-ing decrease in CD 4+ cells in comparison with PBL and LNL.TIL produced lower levels of IFN-γand IL-2upon mitogen PHA and phorbol ester stimulation,while PBL produced high leve ls of TNF-αand IFN-γbut low levels of IL-2.Under the same experimental cond itions,LNL produced high levels of TNF-αand IL-2but low levels of IFN-γ.TIL produced lower levels of TNF-αand IFN-γthan PBL and LNL did.[Conclu-sion]O ur results indicate that the TIL,LNL and PBL from colorectal cancer patients hav e different pat-terns of activities.PBL maybe an ideal origin of CTL precursor cell in the adoptive immunity treatment for colorectal cancer.
