摘 要:目的 :本实验利用基因芯片技术研究缺氧处理后鼠脑星形胶质细胞基因表达谱的变化 ,以深入认识星形胶质细胞在缺氧处理后基因表达变化规律 ,为进一步全面认识神经胶质细胞在缺血情况下基因变化规律以及神经胶质细胞与神经元在损伤条件下相互依存关系提供实验依据和理论基础。方法 :用TRIzol试剂经过多个步骤提取各组制备好的星形胶质细胞样本总RNA ;利用基因芯片技术获取缺氧处理后鼠脑星形胶质细胞的基因表达谱 ;利用GeneOntologyConsortium分析系统、GeneCards数据库和Pubmed检索系统对基因表达谱进行初步分析。结果 :基因表达谱 :在基因芯片上测定了 4 0 96个点 ,共有差异表达基因 15 3个 ,其中 ,已知差异表达基因 4 5个 ,未知差异表达基因 10 8个 (EST片段 ) ;己知差异表达基因 :在 4 5个已知差异表达基因中 ,表达下调的基因有 2 2个 ,表达上调的基因有 2 3个 ,未见报道的与缺氧处理相关基因 2 8个 ,已报道的有 17个 ;已知差异表达基因功能聚类 :共 8类。结论 :本实验获得鼠脑星形胶质细胞缺氧处理后的基因表达谱 ;首次发现 2 8个与缺氧相关的未见报道基因 ;初步明确已知差异表达基因在缺氧处理后的基本变化规律。Objective:To screen the differential expressed genes induced by hypoxia in cultured rat cerebral astrocytes using microarray technology.Methods:Primary cultured cerebral astrocytes (40 days) were exposed to hypoxia caused by mixed gases composing 95% N 2 + 5% CO 2. These cells were collected and total RNA samples were gained by using Trizol extract solution, cDNA microarray chips comprising 4096 clones were employed to identified the effect of hypoxia on gene expression profile in cultured astrocytes. The obtained results were analyzed by using Gene Ontology Consortium analysis system, Gene Cards database and Pubmed Index system.Results:Gene expression profile: There are differentia expression gene 153 points in 4096 measured points, of these ,differentia expression gene known 45points. differentia expression gene unknown 108 points(EST segment);Known differentia expression gene: in the 45 points known differentia expression gene, 22 points were down-regulated,23 points were up-regulated. 28 genes relating to hypoxia have not reported. 17 genes have reported; Function of differentia expression gene known: There are 8 kinds. There are metablism 30 points, signal transmission 4 points, cell growth 4 points, structure protein 2 points,transport 2 points, cell cycle 1 points, cell adhesion 1 point, transmission of never impulse 1 point.Conclusions:The experiment achieve gene expression profile of cultured rat cereberal astrocytes following exposured to hypoxia;28 genes relatd to hypoxia were reported for the first time; The basic rule of differential genes expression during hypoxia in cultured rat cerebral astricytes has been discovered primaryly.
