摘  要:目的 :研究苦参碱诱导人肝癌SMMC 772 1细胞分化过程中对其侵袭转移能力的抑制作用。方法 :SMMC 772 1细胞体外培养 ,0 5 g/L、0 8g/L、1 0 g/L浓度的苦参碱分别处理细胞 ,通过生长曲线测定、细胞形态观察、细胞粘附试验、运动试验、侵袭试验全面观察苦参碱对该细胞株的生长增殖、形态变化、侵袭转移等各方面的诱导分化作用。结果 :经 0 8g/L和 1 0g/L浓度的苦参碱处理后 ,SMMC 772 1细胞的生长增殖明显减缓 ,恶性细胞形态逐渐向成熟细胞分化 ,肿瘤细胞与FN的粘附力、细胞穿膜运动力、对基底膜的降解力等侵袭转移相关指标均受到显著抑制。结论 :苦参碱是一种较好的诱导分化剂 ,不仅抑制细胞的生长增殖 ,诱导细胞形态分化 ,而且从细胞的粘附、运动、降解等多环节全面抑制SMMC 772 1细胞的侵袭转移能力。Objective: To study the inhibition effect on invasion and metastasis related factors in differentiation of SMMC 7721 cells induced by matrine. Method: Human hepatoma cell lines SMMC 7721 were cultured in vitro and treated by 0 5 g/L, 0 8 g/L, 1 0 g/L matrine Then the cell growth curve was protracted, the cell form was observed by light microscope, the invasion and metastasis related factors were measured with cell adhesion test, cell migration test and cell invasion test The data was collected and analyzed with statistical methods to evaluate roundly the inducing differentiation function of matrine Result: Treated by 0 8 g/L, 1 0 g/L matrine, the proliferation of SMMC 7721 was inhibited markedly, and the malignant cell form tended to normal differentiation It was also found that matrine could inhibit cell invasion and metastasis related factors, including adhesive ability to FN, migrative ability through the polycarbonate filter, and invasive ability through matrigel Conclusion: Matrine is a good differentiation inducer, it can not only inhibit the cell proliferation and induce the cell form differentiation, but also significantly inhibit the invasion and metastasis ability of SMMC 7721 through several steps, involved cell adhesion, migration and invasion
