检测胃癌患者外周血微转移端粒酶hTERT-mRNA的意义

摘　　要：目的　探讨检测胃癌血液微转移的新方法 ,评价端粒酶hTERT—mRNA作为检测微转移标志物的价值。方法　提取4 2例胃癌病人外周血有核细胞RNA ,采用RT -PCR方法检测其端粒酶hTERT -mRNA的表达 ,半定量分析 ,并与 2 5名健康者作对照。结果　 4 2例胃癌病人外周血端粒酶hTERT—mRNA的表达显著高于 2 5例健康者 ;Ⅰ期癌与Ⅱ～Ⅳ期癌之间存在显著性差异 ;不同细胞学类型胃癌之间无显著性差异 ;不同病理学类型的胃癌之间不存在显著性差异。结论　RT -PCR检测外周血端粒酶hTERT -mRNA可用来观察胃癌微转移 ,半定量分析更具价值。Objective To searchfor anewmethodindetecting bloodmicrometastasisof gastric cancer,andtO evaluatethe role oftelomerase hTERT-mRNA as a marker for this detection.Method s Nucleated cell RNA in peripheral blood of 42 gastric cancer patients was extracted.telomerase hTERT-mRNA expression Was measured with RT-PCR method and half quantitatively analysed,then mmpared with that from 25 healthy volunteers.Results TelomeraSe hTERT-mRNA expression in nucleated blcod cells from 42 gaStric cancer patients WaS markedly higher than that in healthy volunteers.There WaS marked difference between gaStric e&n.eer in stageⅠand stage Ⅱto Ⅳ.There WaS no marked difference between gastric cancers of different cellular type,nor between gastric cancers with different pathological type.Conclusion Blood micrometastasis of gastric cancer can be identified through detecting telomerase hTERT-mRNA expressionwithRT-PCRmethod,half quantitative analysisismore valuable.
