摘 要:背景与目的:榄香烯是从姜科植物莪术中提取的抗肿瘤药物,我们应用榄香烯治疗神经胶质瘤取得了较为显著的疗效,本研究探讨榄香烯的抗胶质瘤细胞增殖和诱导凋亡作用。方法:3H-TdR掺入法检测榄香烯对鼠源性C6胶质瘤细胞及人源性SHG-44胶质瘤细胞的体外增殖抑制效应,Hoechst33258/PI荧光染色法观察细胞形态学改变,流式细胞仪和DNA凝胶电泳检测细胞凋亡。结果:榄香烯能有效抑制C6/SHG-44胶质瘤细胞的恶性增殖,3H-TdR掺入法检测榄香烯处理不同时间(1~4天)的IC50,C6细胞为7.33~11.02mg/L,SHG-44细胞为13.29~27.16mg/L。在相同药物浓度下,C6细胞较SHG-44细胞敏感。经榄香烯作用后,C6/SHG-44胶质瘤细胞核固缩、边集,核浆比例减小并见有凋亡小体形成,DNA直方图观察到特征性亚倍体峰(Ap峰),DNA凝胶电泳出现有明显的梯状条带。结论:榄香烯有显著的抗胶质瘤细胞增殖作用,并能诱导胶质瘤细胞凋亡。BACKGROUND &OBJECTIVE:Elemene,isolated from the Chinese medicinal herb Rhizoma Zedo ariae,was shown to exhibit antitumo r activity.Our previous studies show ed that elemene had a markedly antineoplastic activity on g lioma.This study was desig ned to investig a te the proliferation inhibitory effect and the apoptosis-inducing activity o f elemene on g lioma cells.METHODS :The effects of elemene on cell prolif eration were studied in vitro by using 3 H-TdR incorporation.The morpholog ical alterations were confirmed by Hoechst 33258/PI s taining .The apoptosis was evaluate d by flow cytometry analysis and ag arose g el electrophoresis.RESULTS :Elemene exhibited a marked antiproliferative effect on rat g lioma cell C6and hum an g lioma cell SHG-44.The fifty percent inhib ition concentration(IC 50 )of elemene ag ainst g lioma cell lines at different time p oints(D1-D4)by 3 H-TdR incorporation was C67.33-11.02mg /L,SHG-4413.29-27.16mg /L.At the same concentration,human g lioma cell line SHG -44was fou nd to be less sensitive to elemene compared to rodent cell line C6.The c haracteristic nucleolus alternations under fluorescent microscope included co ndensation of chromatin arrang ed un der the nuclear membrane and apoptotic bodi es,with a low nuclear /cytoplasmic r atio.In flow cytometry analysis,a typica l subdiploid peak before Phase G 0 /G 1 (apoptotic peak)was detected in DNA frequency distribution histog rams.Also the apoptosis in g lioma cells was con firmed by DNA ladder formation on g el electrophoresis.CONCLUSION:Elemene exhibited a marked antiprol iferative effect on g lioma cells ,and it could i nduce apoptosis in vitro.
