摘  要:目的观察贫血小板血浆(platelet-poor plasma,PPP)和根面脱矿单独及联合应用对人牙龈成纤维细胞(human gingival fibroblasts,HGFs),在病变牙根面的附着及增殖的影响。方法体外培养牙龈成纤维细胞;应用梯度密度二步离心法从人新鲜全血中获取PPP;选取因重度牙周病拔除的病牙制备成病变牙根片。经PPP和EDTA(24%,pH=6.7)分别和联合处理的病变根片的三组作为实验组,未处理的病变根片作为对照组,共分成四组,之后将培养5至8代的细胞分别接种到各组根片上,分别通过细胞计数法、四唑盐比色法[3-(4,5-dimethylthiazo1-2-y1)-2,5-diphenyl tetrazolium bromide,MTT]检测牙龈细胞在不同处理病变牙根表面的附着和增殖。结果(1)细胞计数结果:与未处理组相比,PPP组及根面脱矿组均能促进人牙龈细胞对病变根面的附着(P【0.05),二者联合处理促细胞附着效果明显增强(P【0.01),单独经PPP处理和根面脱矿处理两者比较,细胞对病变根面的附着无明显差异(P】0.05),与单独经PPP处理和根面脱矿处理相比,PPP和根面脱矿联合处理促细胞附着效果明显增强(P【0.01);(2)MTT结果显示:未处理组中人牙龈细胞体外培养48h与培养24h相比,细胞在病变根片上的无增殖(P【0.05);单独PPP或脱矿处理组中人牙龈细胞体外培养48h与培养24h相比,细胞在病变根片上的附着及增殖增加,但无统计学意义(P】0.05),PPP和根面脱矿联合处理组中细胞在病变根片上的附着及增殖明显增加(P【0.05)。结论PPP和根面脱矿单独和联合处理病变根片能明显加强三种细胞的附着和增殖,而PPP和根面脱矿联合处理还有显著的促细胞增值效应。这些崭新的根面处理方式为在牙周病治疗中促牙周组织再生的应用奠定了理论和实验基础。Objectives The purpose of this investigation was to determine whether it is possible to enhance human gingival fibroblasts attachment and proliferation to diseased root surfaces of human diseased root surface treated with EDTA and freshly prepared platelet-poor plasma (PPP) from human whole blood. Methods Human gingival fibroblasts (HGFs) were grown from tissue explants and cells between the 5th and 8th passage in culture were used. Human whole blood obtained from healthy subjects was collected in tubes containing an anticoagulant (acid-citrate-dextrose) by gradient density centrifugation. Periodontal-affected teeth were collected and sectioned to prepare the 4mm×4mm×1mm diseased root slices. There were four groups involving in the experiment:the untreated root slices, the root slices treated with EDTA (24%,pH=6.7), the root slices treated with PPP alone and treated with EDTA and PPP. The cell attachment were assessed by cell counting and the proliferation were assessed using MTT assay. Results Cell attachment and proliferation were significantly increased in experiment group contrast to control group. Conclusions Pretreatment of diseased human root surfaces with freshly prepared PPP or EDTA may enhance the attachment and proliferation of human gingival fibroblasts. This treatment may used as a strategy for treating periodontal defects.
