摘 要:目的:研究表皮生长因子(EGF)对胆囊癌细胞GBC-SD生物动力学,及对uPA,MMP-2,MMP-9,COX-2和NF-κB的mRNA及蛋白表达的影响。方法:通过侵袭、黏附和增殖实验观察在EGF作用下胆囊癌细胞株GBC-SD生物动力学变化;使用RT-PCR和Western Blot检测胆囊癌细胞株GBC-SD的uPA,MMP-2,MMP-9,COX-2和NF-κB的mRNA和蛋白表达,以及在NF-κB抑制物PDTC作用下上述指标的变化。结果:EGF能够显著提高胆囊癌细胞株GBC-SD的侵袭力,但对增殖及黏附力无显著影响;除MMP-2外,随着EGF浓度的增加,uPA,MMP-9,COX-2和NF-κB的mRNA和蛋白表达显著增强;PDTC可以抑制EGF诱导的上述基因mRNA和蛋白水平变化,但MMP-2未见显著变化。结论:EGF可以通过NF-κB途径上调uPA,MMP-9,COX-2和NF-κB表达,促进胆囊癌细胞的侵袭和转移。Objective:To study the effects of growth factor (EGF) on biodynamics of carcinoma of gallbladder cell line GBC-SD, and the influence on mRNA and protein expression of uPA,MMP-2, MMP-9,COX-2 and NF-κB. Methods:The effects of EGF on biodynamics of carcinoma of gallbladder cell line GBC-SD were investigated by invasion, adhesion and proliferation experiments; mRNA and protein expressions of uPA,MMP-2, MMP-9,COX-2 and NF-κB with/without the intervention of pyrrolidine dithiocarbamate(PDTC), a NF-κB inhibitor, were evaluated by means of RT-PCR and Western Blot. Results:EGF could significantly increase the invasion of GBC-SD but its proliferation and adhesion. With the increase of EGF concentration, the mRNA and protein expressions of uPA,MMP-9,COX-2 and NF-κB were obviously enhanced but MMP-2. Moreover,PDTC could inhibit the expressions change of above-mentioned genes but MMP-2. Conclusion:EGF can enhance the expressions of uPA,MMP-9,COX-2 and accelerate the invasion and metastasis of GBC-SD throngh NF-κB pathng way.
