美解眠致痫大鼠海马组织损伤及其NO、SOD的变化

摘  要:目的 观察癫痫大鼠海马组织病理变化及海马一氧化氮 (NO)、超氧化物歧化酶 (SOD)的变化 ,并探讨海马组织损伤与NO、SOD的关系。方法 通过美解眠制成癫痫发作动物模型 ,分别取发作 5min、30min及注药 6h、12h、2 4h的海马组织 ,经典病理组织切片及硝酸还原酶法、黄嘌呤氧化酶法测定NO、SOD含量 ,光镜观察海马神经元丢失情况 ,TUNEL法阳性凋亡细胞计数。结果 ①海马组织在癫痫发作 5min起即有轻度细胞水肿 ;发作 30min病变则水肿明显 ;6h组海马锥体细胞和颗粒细胞变性、坏死 ,开始出现凋亡细胞 ;2 4h时凋亡细胞计数达最大 ;2 4h残存细胞最少。②海马组织NO在发作 5min迅速升高 ,30min组仍高于正常对照组 ,6h以后NO与正常对照组无显著差别 ;海马组织SOD于发作 5min时显著低于正常对照组 ;6h时仍与对照组存在差别 ;12h以后与对照组差别无统计学意义。结论 癫痫发作可导致海马组织严重损伤 ,并随时间推移而加重 。Objective To observe the pathological changes and the contents of superoxide in hippocampal tissue dismustase and nitric oxide after seizure induced by megimide,and study the relationship between them. Methods An animal model of epilepsy was made by megimide 25 mg/kg ip. NO and SOD were measured by classic histopathological method and by TUNEL. Results ①It was found that the peak time the neurons left after seizure was at 24 h group. The apoptosis neurons increased at 6 h group and at 24 h group reached the maximum in hippocampus.② the SOD was lowest in 5 min group,and was still lower in 30 min group and 6 h group,it became normal in 12 h and 24 h ones; the nitric oxide increased greatly in 5 min group, moderate high in 30 min group in hippocampus,but not in 6 h group and 12 h group.Conclusion The damage induced by seizure has some relation with the risen NO and free radical in hippocampus.
