摘  要:目的 :观察辐射对 IM- 9细胞间期核内第 9,2 2号染色体三维空间结构的影响 ,探讨 t(9;2 2 ) (q34;q12 )染色体易位的形成机制 ,从生物体视学角度探讨慢性髓系白血病发病机制。方法 :运用激光共聚焦显微镜技术和荧光原位杂交方法 ,分析人淋巴细胞株 IM- 9间期核内第 9,2 2号染色体的体积和表面积在细胞周期中的变化 ,观察辐射对其产生的影响并同 K5 6 2细胞相比较。结果 :IM- 9细胞间期核内第 9或 2 2号染色体的体积在细胞周期的同一时相内同 K5 6 2细胞无明显差别 (P>0 .0 5 ) ,放射对其也无影响 ;但放射可以使第 9或 2 2号染色体的表面积明显增加 ,并且在 S期和 G2 / M期明显大于 G1 期 (P<0 .0 5 ) ;这一特征同 K5 6 2细胞的表现是相一致的。结论 :辐射可以改变 IM- 9细胞染色体的三维空间结构并使其更具有慢性髓系白血病细胞的特征 ,这种改变对于慢性髓系白血病的发病可能具有重要意义。Objective:To explore the mechanism of the formation of t (9; 22) by observing irradiation influence on the three dimensional structure of the chromosome 9 and 22 in IM 9 cell interphase nucleous, in order to elucidate the pathogenesis of chronic myeloid leukemia.Methods:In combination of confocal laser scanning microscope and fluorescence in situ hybridization, the dynamic changes of the volume and surface area of the chromosomes 9 and 22 in the interphase nucleous of IM 9, a human lymphocyte cell line, were investigated, the irradiation influence on them was obvserved , and was compared with that of K562 cells.Results:No difference of chromosomes 9 or chromosome 22 volume was noted between the IM 9 and K562 cells ( P >0.05) in the same cell cycle phase, and the volumes were not influenced by irradiation. However, irradiation increased the surface area of chromosomes 9 and 22 markedly, and the surface areas in S or G 2/M phase were much greater than that in G 1 phase ( P <0.05) in both IM 9 and K562 cells.Conclusion:Irradiation could change the three dimensional structure of chromosomes 9 and 22 in IM 9 cells and endow it with the character similar to K562 cells. These changes might act an important role in the pathogenesis of chronic myeloid leukemia.
