膀胱癌细胞凋亡与多药耐药的实验研究

摘  要:目的 研究膀胱癌细胞凋亡与膀胱癌多药耐药的关系。方法 用电镜观察几种临床常用的化疗药物阿霉素、顺铂和丝裂霉素在体外诱导膀胱癌细胞的凋亡的形态学特征 ,免疫组化测定pgp糖蛋白的表达 ,应用MTT法检测阿霉素、顺铂和丝裂霉素的杀伤率。结果  16例患者中 9例有效地诱导出细胞凋亡 ,7例未诱导出凋亡 ,电镜观察显示 ,患者的膀胱癌细胞对化疗药物阿霉素、顺铂和丝裂霉素敏感时 ,其细胞呈特异性的凋亡形态学特征改变 :染色质聚集于核膜呈块状 ,扩张的线粒体 ,嵴移位膨胀 ,凋亡细胞被周围细胞吸收。体外检测显示耐药患者肿瘤细胞抑制率低于 3 0 %。结论 研究结果提示多药耐药的产生与膀胱癌细胞凋亡受到抑制有关。Objective To study the relation between bladder cancer cell apoptosis and Multidrug-resistance.Methods lnduced apoptotic of bladder cancer cell was analyzed by electron-microscorpic.Kill effect of ADR?CDDP and MMC on cultured bladder cancer cell were detected by MTT method.Results The nine of sixteen patients`bladder cancer cell showde the characteristic change of apoposis morphology by Treated ADR?CDDPand MMC,confluent dense-cone chromatin was attached to nuclear envelope,moreover,lots of apoptosis cell`s mitochondria swell,mitochondria cristae modified,apoptosis cell was being absorb by around its cell.Other seven people were not showed one .lnhibition rate of multidrug-resistance patents lower 30% by MTT in vitro tumor cell detection.Conclusions According to results of research could infer the multidrug -resistance of bladder cancer cell was related to inhibition of bladder cancer cell apoptosis.
