摘 要:目的:探讨中药红景天有效单体成分红景天甙抑制乙醛刺激的体外培养大鼠肝星状细胞(hepatic stellate cell,risc)增殖的机制。方法:用免疫细胞化学染色法检测HSC内增殖细胞核抗原(Proliferating Cell Nuclear Antigen,PC-NA)的表达,流式细胞仪分析红景天甙作用于乙醛刺激的HSC后细胞周期的变化。结果:红景天甙组HSC内PCNA 表达明显低于乙醛组(0.34±0.07 Va 0.62±0.04),P<0.05;红景天甙可明显抑制乙醛刺激HSC由G1-S期转化,表现为红景天甙组GD/G1期细胞百分率较乙醛组显著增加(59.25±4.55 Vs 36.12±3.01),P<0.05;S期细胞百分率显著降低(30.34±4.35Vs 51.26±2.84),P<0.05。结论:红景天甙抑制HSC增殖可能与其阻滞HSC细胞周期有关。Objective: To explore the growth suppression mechanisms of Chinese medicine salidr os ide on hepatic stellate cells (HSC) in rats stimulated by acetaldehyde. Method: Immunocytochemical stain was employed to detect the expression of proliferating cell nuclear antigen. (PCNA) in HSC. Result; The expression of PCNA in salidrosicde group was significant lower than that in acetaldehyde group (0.34 ±0.07 Vs 0.62 ±0.04, P<0.05). Compared with that in acetaldehyde group, the percentage of G0/G1 cells m saliroside grup significantly increased (59.25 ±4.55 Vs 36.12± 3.01, P<0.05) but the percentage of S cells decreased (30.34±4.35 Vs 51.26±2.84, P<0.05), which indicated that salidroside can significantly inhibit the transition of acetaldehyde - stimulated HSC from G1 phase to S. Conclusion: The inhibitory effect of saidroside on HSC proliferation might be connected with the mechanism of salidroside inhibiting the cell cycle of HSC.
