摘 要:目的 :探讨三种激光能量的光动力效应对体外培养的人Tenon囊成纤维细胞增殖的抑制作用。方法 :在体外培养的人Tenon囊成纤维细胞培养液中依次加入光敏剂BPD ,使其终浓度分别为 3 9 0 62 5ng/ml、 78 12 5ng/ml、15 6 2 5ng/ml、 3 12 5ng/ml、 62 5ng/ml、 12 5 0ng/ml、 2 5 0 0ng/ml,分别给予 1 2J/cm2 、 2 4J/cm2 和 3 6J/cm2 半导体激光照射 ;另设空白对照组 ,未加任何处理。MTT法测各孔光吸收值 (OD值 ) ,分别计算各组抑制率 ,并采用SPSS软件 ( 10 0 )进行统计学分析。结果 :三种照光能量的IC50 分别为 192 0 68ng/ml、 114 14 2ng/ml和 91 970ng/ml。经单因素方差分析及用LSD法进行各两组间的多重比较 ,三种照光能量组内 ,光动力效应组均与空白对照组间有统计学差异 ;照光能量为 1 2J/cm2 时 ,各组成纤维细胞抑制率分别为 11 62 %、 2 6 13 %、 47 68%、 74 0 5 %、 88 0 8%、 90 76%和 92 5 4% ,;照光能量为 2 4J/cm2 时 ,各组抑制率为12 5 %、 41 6%、 66%、 87 1%、 90 3 %、 91 0 %和 93 3 % ;照光能量为 3 6J/cm2 时 ,各组抑制率为 2 1 97%、 48 5 4%、78 3 7%、 86 5 2 %、 91 85 %、 91 85 %和 94 85 %。结论Objective:To investigate the different photodynamic effects of three levels of laser energy on the human Tenon capsule fibroblasts in vitro. Methods:Cultured human Tenon capsule fibroblasts seeded in three 96-well plates were given the photosensitizer benzoporphyrin derivative monoacid ring A(BPD) in 7 different concentrations.The end concentration of every group were 39 0625ng/ml,78 125ng/ml,156 25ng/ml,312 5ng/ml,625ng/ml,1250ng/ml and 2500ng/ml,respectively.Fifteen minutes later,each plate was exposed to 689nm diode laser with different energy of 1 2J/cm 2?2 4J/cm 2和3 6J/cm 2,respectively.The control group was given no treatment.The OD values were detected by MTT method after 24 hours and the inhibition rate of each group was calculated. Results:The IC 50 of three laser energy was 192 068ng/ml,114 142ng/ml and 91 970ng/ml,respectively.There were statistical significances between photodynamic effect groups and control group.The relative inhibitory rates were 11 62%,26 13%,47 68%,74 05%,88 08%,90 76% and 92 54% with laser energy of 1 2J/cm 2,12 5%,41 6%,66%,87 1%,90 3%,91 0% and 93 3% with laser energy of 2 4J/cm 2,and 21 97%,48 54%,78 37%,86 52%,91 85%,91 85% and 94 85% with laser energy of 3 6J/cm 2. Conclusions:Photodynamic effect can inhibit the proliferation of human Tenon capsule fibroblasts in vitro.The inhibitory rate appears to be dependent on the concentration of the photosensitizer and the energy of laser.The combination of BPD 156 25ng/ml and laser energy 3 6J/cm 2 may achieve the best effect.
