中药地椒提取物的抗肿瘤作用及对小鼠免疫功能的影响

摘　　要：目的　探讨中药地椒提取物的抗肿瘤作用及其对小鼠免疫功能的影响。方法　从野生地椒中提取挥发油及乙醇提取物并制成受试溶液 ,以荷S180瘤小鼠为模型 ,用不同浓度的地椒挥发油和乙醇提取物受试溶液给小鼠灌胃 ,连续给药 9d后 ,测算地椒提取物组、正常组、阴性对照组和环磷酰胺组各小鼠肿瘤、脾脏及胸腺的重量 ,计算抑瘤率、脾指数和胸腺指数。结果　地椒乙醇提取物高浓度组 (40g生药·kg- 1·d- 1)抑瘤率为 51.5% (P <0 .0 1) ,低浓度组 (2 0g生药·kg- 1·d- 1)抑瘤率为3 6.4% (P <0 .0 5) ;地椒挥发油高浓度组 (40g生药·kg- 1·d- 1)和低浓度组 (2 0g生药·kg- 1·d- 1)抑瘤率均为 3 9.4% (P <0 .0 5)。小鼠荷瘤后出现免疫指标的异常 ,表现为脾指数的升高和胸腺指数的下降。地椒乙醇提取物高浓度组可使荷瘤小鼠的脾指数降低并趋向正常 ,胸腺指数则介于阴性对照组和环磷酰胺组之间 ,其他地椒提取物组也有类似作用。结论　地椒挥发油及乙醇提取物均能抑制小鼠体内移植性肿瘤的生长 ,乙醇提取物的抗肿瘤作用更明显 ,并且表现为药物浓度依赖性。此外 。ObjectiveTo study the anti-tumor and immunologica l effect of extracts from Thymus quinquecostatus Celak on mice transplanted S 180 tumor cells. MethodsDifferent doses of volatile oil and alcohol extracted substances from Thymus quinquecostatus Celak were given to mice bearing S180 tumor for 9 days. Tumor inhibition rates and coefficients of spleen and thymus were determined. ResultsTumor inhibition rates of the groups with alcohol extracts(40 g crude drug·kg -1 ·d -1 and 20 g crude drug·kg -1 ·d -1 )were 51.5%(P <0.01) and 36.4%(P <0.05)respectively,and those of the groups with volatile oil(40 g crude drug ·kg -1 ·d -1 and 20 g crude drug·kg -1 ·d -1 )were both 39 .4%(P <0.05). ConclusionThe extracts from Thymus quinqueco status Celak have anti-tumor activities. The coefficient of spleen in group w ith alcohol extracts(40 g crude drug·kg -1 ·d -1 )was close to norma l value, and its coefficient of thymus was between that of the negative control group and the group with cyclophosphamide(0.02 g·kg -1 ·d -1 ). The anti-tumor activity of the alcohol extracts was significantly higher than that of the control group and the tumor inhibition rate was depending on drug concent ration. Depending on index of immunity,the extracts from Thymus quinquecostat us Celak may have some influences on immunity.
