摘　　要：目的　了解急性髓细胞白血病 (AML)M5亚型细胞诱导T细胞的TCRVβ亚家族限制性利用和克隆性增殖情况。方法　采用混合淋巴细胞 /肿瘤细胞培养 (MLTC)方法 ,用AML M5细胞体外诱导供者外周血和脐血的T细胞增殖 ,用RT PCR扩增经MLTC前后获得细胞的 2 4个TCRVβ亚家族基因的互补决定区 (CDR3) ,了解病人各Vβ亚家族的利用情况。并经基因扫描分析确定M5细胞诱导的T细胞克隆性特点。结果　脐血和供者外周血分别表达 13和 5个TCRVβ亚家族T细胞 ,经MLTC培养初期 ,则表达全部 2 4个Vβ亚家族 ,随着诱导时间的增加 ,诱导T细胞所表达的Vβ亚家族逐渐减少。基因扫描显示AML M5细胞诱导后的T细胞出现寡克隆和双克隆增殖改变 ,主要为Vβ2 1的克隆性增殖T细胞。结论　AML M5细胞可诱导TCRVβT细胞克隆性增殖 ,这可能是对AML M5细胞相关抗原的特异细胞免疫应答的结果。Objective To investigate the restricted usage and clonal expansion of TCR Vβ subfamily T cells induced by AML-M_5 cells. Methods The mononuclear cells(MNC) from donor peripheral blood or cord blood were amplified by mixed lymphocyte and tumor culture(MLTC), induced by AML-M_5 cells from 2 patients respectively. The complementarity determining region 3(CDR3) of TCR β with variable region genes was amplified in T cells from the donor or cord blood before and after MLTC using RT-PCR, to detect the usage feature of TCR Vβ subfamilies. The positive products were further analyzed to identify the clonality of T cells by genescan. Results Only 13 or 5 TCR Vβ subfamilies were expressed in T cells from cord blood or donor, however, 24 TCR Vβ subfamily T cells could be detected in T cells from both cord blood and donor at early stages after MLTC, whereas the number of expressed TCR Vβ subfamilies was gradually reduced with prolonging the time of culture. Oligoclonal and biclonal T cell expansion predominated in Vβ21 could be identified in T cells from cord blood and donor induced by AML-M_5 cells by genescan analysis. Conclusion Clonal expansion of TCR Vβ subfamily T cells from cord blood or donor could be induced by AML-M_5 cells, which may be a specific immune response to AML-M_5 cell associated antigen.
