乳腺癌细胞DNA定量分析

摘  要:应用流式细胞术检测了乳腺良恶性病变的DNA含量及倍体水平.20例乳腺良性病变均为二倍体,90例乳腺癌中异倍体肿瘤75例(83.3%),二倍体肿瘤15例(16.7%);Ⅰ,Ⅱ,Ⅲ,各级不同恶性程度肿瘤的DNA指数(DI)分别为1.23±0.14,1.32±0.18和1.61±0.30;5年存活组与死亡组DI值分别为1.21±0.19和1.53±0.28.细胞增殖周期分析表明二倍体肿瘤和异倍体肿瘤分别以G_0/G_1期细胞和S+G_2 M期细胞占的比例较高.本文未发现肿瘤体积及组织类型(WHO)与DI值的相关性.提示检测DNA含量和细胞增殖周期,对评估乳腺癌的恶性程度及其生物学行为有重要价值.DNA ploidy of cancer and benign lesions of the breast were studied by flow cytometry. Diploidy was found in all of the 20 cases of benign lesions. In the 90 cases of breast cancer, aneuploidy was found in 75(83.3%)and diploidy in 15 (16.7%). The DNA indices(DI)were 1.23±0.14,1.32±0.18 and 1.61±0.30 in the histological malignant grades of Ⅰ, Ⅱand Ⅲ,and 1.21±0.19 and 1.53±0.28 in the 5-year-survival and dead groups respectively. It was found that the diploid and aneuploid tumors were larger at the phases of G_0/G_1 and S+G_2M in their cell cycle respectively. The data were statistically significant. In this study no relationship was found between DI and the size of the tumor and between the DI and the histological classification as laid down by WHO. It is believed that the cell cycle and DI determination of breast cancer might be valuable in representing the grade of malignancy and biological behavior of tumors.
