硒对中期保存的兔角膜内皮细胞的影响

摘　　要：目的　探讨硒对中期保存的兔角膜内皮细胞的保护作用及机制。方法　前房内分别注入M -K液 ,以及含有 3 μmol/L、5μmol/L、8μmol/L亚硒酸钠的M -K液后 ,全眼球保存兔角膜。在保存 1、3、5d后 ,分别行内皮细胞谷胱甘肽过氧化物酶 (GSH Px)活力检测、角膜内皮细胞活性染色、角膜内皮显微镜检查。结果　保存后实验组GSH Px活力明显高于对照组。保存 3d及 5d后实验组内皮细胞存活率高于对照组。内皮显微镜显示 ,保存后的六边形细胞百分率各实验组均较对照组高。保存 5d后实验组细胞平均密度高于对照组。结论　在角膜中期保存液中加入一定剂量的硒 ,能够提高内皮细胞的抗氧化能力 ,延长角膜的保存时间。Objective To investigate the protective effec ts of selenium on endothelial cells of mid-term storaged corneas in the rabbit. Methods Rabbit corneas were stored differed in the irriga tions of anterior chamber with M-K liquid,M-K liquid with sodium selenite (Na 2SeO 3) of 3 μmol/L,5 μmol/L and 8 μmol/L.Enucleation was performed after 1 day,3 days and 5 days respectively.The activity of GSH-Px in endothelial cell s was estimated.Endothelial cells were actively stained by trypan blue.Morpholog ical study was performed by specular microscope and electron microscope.Results The activity of GSH-Px raised compared with the group without selenium.The vital staining showed the endothelium survival rate w as higher after 3 and 5 days.The percentage of the endothelial hexagonal cell wa s also higher,which is showed by specular microscope.The average cell density w ith selenium was higher after 5 days.Under electronic microscope,the ultrastruct ural damages of corneal endothelial cells without selenium were more severe.The consecution of cell membrane was broken.Vacuoles in cytoplasm could be seen an d the construction of organellae was lack of definition. Conclusion The intermediate term storage media with added selenium had significant differences in increasing the vitality of endothelial cells,and could prolong the time of corneal storage.
