摘 要:目的 了解维甲酸纳米粒对兔视网膜是否产生毒性。方法 30只青紫蓝兔的 6 0眼随机分为 5组 ,每组 12眼 ,正常组不注药 ,其余分别向玻璃体腔注入维甲酸纳米粒 5、10、2 0mg·L-1以及BSS(对照组 ) 0 .3mL ,以检眼镜、电生理、光镜和电镜检查观察视网膜变化情况。结果 注药后 7d,光镜检查 2 0mg·L-1组出现神经节细胞的空泡样变 ,电镜及电生理检查未见异常 ,14d时恢复正常 ;至 2 8d ,未发现各浓度的维甲酸纳米粒对视网膜产生毒性作用。结论 玻璃体腔注入浓度≤ 2 0mg·L-1的维甲酸纳米粒 。Objective To evaluate the toxicity of retinoic acid nanoparticles to the retinal tissue.Methods Thirty pigmented rabbits(60 eyes) were divided into five groups randomly.Retinoic acid nanoparticles 0.3mL of 5,10,20mg·L -1 were injected intravitreally in 12 eyes respectively as 3 experimental groups.The control group(12 eyes) were injected intravitreally BSS 0.3mL and the normal group(12 eyes) were injected no drug.After intravitreal injections,all eyes were examined by ophthalmoscopy,flicker electroretinography(F ERG),light microscopy,and transmission electron microscopy on the 7th,14th and 28th day.Results There were bulbar degeneration like changes in retinal ganglion cells of 20mg·L -1 group at the 7th day after intravitreal injections and these changes were disappear at the 14th day and no abnormal changes were found in retina by F ERG,transmission electron microscopy.And no evidence of toxicity was found in retina by opthalmoscopy,F ERG and transmission electron microscopy in every group.Conclusion There was no toxic effect on the retinas by intravitreal injecting retinoic acid nanoparticles equal to or lower than 20mg·L -1 .
