摘 要:目的 对将使用于义齿磁性固位体的两种特殊不锈钢(S_1与S_2)的生物安全性进行初步检测,评价其临床应用的生物相容性。方法 用纯钛及钴铬合金作比较,通过将材料浸提液与体外培养的L-929细胞接触培养,进行细胞的形态学观察及二甲基噻唑二苯基四唑嗅盐(MTT)比色,评价材料对细胞造成的毒性损害程度。结果 细胞的形态学观察显示,随着培养天数的增加,阳性对照组中的脱落细胞、死亡细胞逐渐增多。其余各组细胞大部分继续贴壁生长。细胞密度增加。MTT比色法结果显示,非磁性不锈钢(S_1)和磁性不锈钢(S_2)浸提液对细胞有轻微的毒性,细胞相对增殖率(RGR)降低3%,钴铬合金对细胞表现出1级细胞毒性,RGR降低11%,但4种金属材料组与阴性对照组的吸光度值之间的差异均无统计学意义(P>0.05)。结论 S_1、S_2两种不锈钢仅具有极微弱的细胞毒性,符合临床应用的要求。Objective To investigate the biosafeness of two kinds of special stainless steel materials that will be used in the magnetic retainer in order to evaluate their biocompatibility for clinical application. Methods After mixing the extracting liquid with the L - 929 cells, we evaluated the cytotoxicity of those materials by cell morphological observation and Mil-assay, comparing them with pure titanium and cobalt-chrome alloy. Results The cell morphological observation indicated that with cultivate time increasing, in the positive control, the cells of shedding off or dead cells increased gradually. The cells of the rest groups almost grew while attaching to the plate continuously, and the cell density increased. The result of MTT-assay indicated S, stainless steel and 82 stainless steel showed slight cytotoxicity (the RGR decreased by 3%), Cobalt-Chrome alloy showed class I cytotoxicity (the RGR decreased by 11 % ) . But the statistical analysis result manifested there were not significant differences in the optical densities between metal material groups and the negative control ( P > 0.05). Conclusion The experiment results showed the two kinds of stainless steel (St and 82) only had very faint cytotoxicity, which met the requirement for clinical application. It provided a biological basis for its application in the magnetic attachment.
