摘 要:目的 :研究端粒酶活性在消化道肿瘤组织及其癌旁组织中的表达及其意义 ,并评价TRAP ELISA检测法的应用价值。方法 :用TRAP ELISA方法检测了 112例肿瘤组织 (其中食管癌 12例、胃癌 36例、肝癌 15例、胰腺癌 11例、大肠癌 38例 )及 94例相应癌旁组织的端粒酶活性。结果 :食管癌、胃癌、肝癌、胰腺癌、大肠癌肿瘤组织端粒酶活性阳性率分别为 83.33%、86 .11%、86 .6 7%、90 .90 %和 84 .2 1%。肿瘤组织端粒酶活性明显高于癌旁组织和正常组织 (P <0 .0 0 1) ,端粒酶的活性与肿瘤大小、转移、分化程度、分期均不相关。结论 :端粒酶活性是消化道肿瘤诊断的一个很好的指标 ,而且TRAP ELISA检测法是一种非放射性同位素方法 。Purpose:To investigate the telomerase activity in human digestive tract cancer and its significance and evaluate usefulness of the TRAP ELISA method .Methods:Telomerase activity was examined in 112 tumor specimens, including the following cancers: esophageal(12), gastric(36), hepatic(15), pancreatic (11), colorectal(38) and paracancerous tissues(94) by TRAP ELISA method.Results:The telomerase activity in tumor tissues was significantly higher than in paracancerous and normal tissues ( P <0.001). Positive rates of telomerase activity in esophageal, gastric, hepatic, pancreatic and colorectal cacinomatous tissues were 83.33%, 86.11%, 86.67%, 90.90%, 84.21%, respectively. It was found that telomerase activity was not related to size of tumor , the degree of differentiation, clinical staging and lymph node metastasis. The TRAP ELISA method need not use isotopes, being highly specific and less time consuming.Conclusions:Telomerase activity may be a good tumor marker for detection of digestive cancer. The TRAP ELISA method is non radioisotopic, semi quantitative and a rapid determining method for detection of cancerous telomerase activity.
