摘 要:目的 研究全反式维甲酸 (ATRA)对人肝癌高转移细胞株MHCC97 H表达基质金属蛋白酶 (MMPs)的影响。方法 应用明胶酶谱法检测ATRA对体外培养MHCC97 H细胞分泌MMPs酶谱的影响 ;细胞免疫荧光法检测ATRA对MHCC97 H细胞表达MMP 9的影响 ;利用侵袭小室模型 ,检测ATRA对MHCC97 H细胞体外侵袭能力的影响。结果 MHCC97 H在体外培养时可分泌大量的MMPs ,其中以MMP 9和其活性形式为主。ATRA在体外可显著地抑制MHCC97 H细胞MMPs的表达 ,降低肿瘤细胞穿透人工重组基底膜的能力 ,且在一定的浓度范围内 ,抑制作用与药物浓度呈正相关。结论 抑制肿瘤细胞分泌MMPs的能力 。Objective To investigate the repression of MMPs secretion by all trans retinoic acid (ATRA) in human hepatocellular carcinoma cell strain MHCC97 H.Methods Gelatin zymography and indirect immunofluresencestaining were used to detect the secretion of MMPs of MHCC97 H cells incubated with ATRA at different concentrations.By using the Boyden chamber model,the influence of ATRA on invasion ability of MHCC97 H cells in vitro was determined.Results Abundance of MMPs was secreted by the cultured MHCC97 H cells,in which,MMP 9 and its activated type were dominant.In vitro,ATRA could evidently decrease the secretion of MMPs by MHCC97 H cells,and depress the ability of MHCC97 H cells to penetrate the artificial reconstituted basement membrane.In the definite range of ATRA concentration,the depression effect was dose related.Conclusion Repression of matrix metalloproteinases secretion was related to the mechanism of antitumor effect of ATRA in tumor invasion and metastasis.
