中药拮新康复方对K562/A02细胞的增殖及膜流动性的影响

摘　　要：目的 :利用血清药理学方法研究中药拮新康复方对K5 6 2 /A0 2细胞的增殖及膜流动性的影响。方法 :分别采用MTT法及荧光分光光度法观察经含拮新康中药血清处理后K5 6 2 /A0 2细胞的增殖及膜流动性的改变。结果 :含 10 %拮新康血清 +ADM (5 0 μg/ml)组对K5 6 2 /A0 2细胞的抑制率较 10 %正常大鼠血清 +ADM (5 0 μg/ml)组明显增大 (P <0 .0 5 )。含 10 %拮新康血清 +ADM组逆转K5 6 2 /A0 2细胞的耐药倍数与含异博定 (5 μg/ml)的阳性对照组的逆转耐药倍数大致相等 (P >0 .0 5 )。含 10 %拮新康血清能显著降低K5 6 2 /A0 2细胞膜流动性 (P<0 .0 5 )。结论 :拮新康能抑制K5 6 2 /A0 2细胞的增殖 ,降低细胞膜的流动性。Objective To investigate the effects of Chinese medicine compound Jiexinkang on the proliferation and membrane fluidity of K562/A02 cell by serum pharmacology. Methods MTT method and fluorospectrophotometry were used to observe the alteration of the proliferation and membrane fluidity of K562/A02 cell after treating with rat serum containing Jiexinkang. Results The inhibiting rate on K562/A02 cell of 10% rat serum containing Jiexinkang and adriamycin (50 μg/ml) was significantly higher than that containing 10% normal rat serum and adriamycin (50 μg/ml) ( P <0.05). The ability of 10% rat serum containing Jiexinkang and adriamycin (50 μg/ml) to reverse drug resistance of K562/A02 cell was approximately equal to that of the positive control containing verapamil (5 μg/ml) ( P >0.05). The membrane fluidity of K562/A02 cell was significantly lowered by 10% rat serum containing Jiexinkang( P <0.05).Conclusion Jiexinkang can inhibit the proliferation of K562/A02 cell and lower it's membrane fluidity.
