摘  要:目的 :观察姜黄素对克隆化人肺癌高转移细胞亚系 (PGCL3)生物学行为的影响。方法 :用 6 0 μ mol/ L的姜黄素处理 PGCL 3细胞 ,通过制备生长曲线及软琼脂集落形成能力试验来评价药物对细胞生长的影响 ,以对层粘连蛋白的粘附试验、通过 Boyden小室聚碳酸酯膜的运动试验和降解穿过基底膜胶的侵袭降解试验来评价细胞侵袭能力的改变。结果 :在 6 0μ mol/ L姜黄素作用下 ,PGCL 3细胞的生长速度及软琼脂集落形成能力、以层粘连蛋白为粘附基质的粘附能力、穿过重建基底膜的侵袭降解能力及趋化运动能力均显著降低 (P<0 .0 1或 P<0 .0 5 )。结论 :姜黄素可抑制 PG-CL 3细胞的恶性表型 ,为治疗转移性恶性肿瘤提供了实验依据。Objective:To observe the effects of curcumin on the biological behaviors of a high metastatic human lung cancer subline(PGCL3).Methods:PGCL3 cells were pretreated with 60 μ mol/L curcumin for 3 days in vitro .The influence of the curcumin on cell proliferation were observed by making growth curve and colony forming ability in semisolid agar culture test.The invasiveness of the cells were evaluated by adhesion test to laminin substrate,motility test through the polycarbonate filter membrane and invasion test to degrade and cross matrigel in Boyden chamber.Results:At the concentration of 60 μ mol/L curcumin resulted in significant decrease of the cells proliferation rate,colony forming ability in semisolid agar culture,adhesion to laminin substrate,migration capacity through the polycarbonate filter membrane,and invasion ability through matrigel ( P <0.01 or P <0.05). Conclusion:Curcumin pretreatment can inhibit the malignant phenotype of a coloned metastatic human lung cancer subline,and the study may provide some reliable experimental data for the therapeutic effect of curcumin on tumor invasion and metastasis.
