摘 要:目的 观察烧伤 12 0h内小鼠腹腔巨噬细胞 (MΦ)TNFαmRNA表达的变化及参麦注射液体内应用的影响。 方法 采用 11%TBSAⅢ度烧伤模型 ,腹腔注射参麦液 (2 .5ml·kg-1·d-1,1ml注射液含红参 10 0mg、麦冬 10 0mg)连续 5d ,运用半定量RT -PCR方法测定TNFα的表达。 结果 烧伤组MΦTNFαmRNA的水平明显增高 ,注射组可显著降低烧伤小鼠腹腔MΦTNFαmRNA的水平 (P<0 .0 1) ,并提高烧伤小鼠的存活率 (P <0 .0 5 )。 结论 烧伤小鼠早期死亡的原因可能与TNFα的增高有关。参麦注射液可以降低烧伤小鼠TNFαmRNA表达、提高烧伤小鼠存活率。Objective To explore the effect of Shenmai injection on the expression of TNFα mRNA of peritoneal macrophages in scalded mice. Methods BALB/c mice inflicted with 11% TBSA Ⅲ degree scalding injury were employed as the model. Shenmai injection was composed of 100 mg of red ginseng and 100 mg of lilyturf root in 1 ml injection solution and was administered to mice via pertioneal injection in a dose of 2.5 ml per kilogram of mice body weight per day for 5 days. The expression of TNFα mRNA was determined by semi-quantitative RT-PCR. Results The expression of macrophage TNFα mRNA was significantly increased in the scalded mice, while the expression was obviously lowered after the administration of Shenmai injection(P<0.01) to scalded mice and the survival rate of the scalded mice was thus markedly increased (P<0.05). Conclusion The beneficial effect of Shenmai injection on the scalded mice might be related to its inhibitory effect on the hyperexpression of macrophage TNFα mRNA. Shenmai injection could lower down the expression of TNFα in scalded mice and raise the survival rate of scalded mice.
