摘 要:为了探讨慢性髓性白血病(CML)易感性与HLA-DRB1等位基因多态性之间的关联性,试图找出慢性髓性白血病的易感基因。采用序列特异性引物聚合酶链反应(PCR-SSP)DNA分型技术对48例CML患者和105例健康人对照组进行了HLA-DRB1基因分型。结果显示,CML与HLA-DR4基因关联,基因频率为22.9%,RR=5.076,x^2=17.88,P<0.01;其它等位基困频率在CML组与对照组间差异无显著性。提示在河南汉族人群中,HLA-DR4与CML有关联。In order to explore the association between HLA-DRBl alleles polymorphism and susceptibility to chronic myelogenous leukemia (CML) in Chinese Henan Han people and find the susceptible genes of CML, 105 normal subjects and 48 patients of Henan Han Nationality with CML were examined for HLA-DRBl genotyping analysis by polymerase chain reaction with sequence specific primers (PCR-SSP). Compared with normal controls, the frequency of HLA-DR4 was significantly increased in the patient group, with a relative risk (RR) of 5.076, x2 =17.88, P<0.01 and with an etiological fraction (EF) of 0.18, while the frequencies of other DRB1 alleles were not significantly increased in the CML group. These results showed that susceptibility to CML is strongly associated with the HLA-DR4 gene.
