人结肠腺癌裸鼠移植瘤模型的建立和生物学特性及放射免疫显象的研究

摘　　要：我院自1989年2月建立的人结肠癌裸鼠移植瘤模型(CTNM),至今已传27代,传代裸鼠共80只,移植成功率100％。对移植瘤和瘤源作系统的生物学特性的研究,测定其肿瘤组织与宿主血液中的癌胚抗原(CEA)值。并应用免疫组化法观察肿瘤组织中CEA分布。结果显示,移植瘤的形态和功能特性与瘤源基本一致。对荷瘤裸鼠放射免疫显象结果表明:经腹腔内注射^(131)I-CEA-McAb 72h后,肿瘤显象满意,瘤/肠(T/NT)值为4.3,标记抗体能在肿瘤组织中浓聚。A human colonic adenooarcinoma model in nude mice has been established since Feb. 1989 in our hospital. So far,the transplanted tumors have been passaged in 80 mice for 27 generations and showed 100% of take rate. Specimens of tumor tissue and blood samples taken from the hosts were subjected to OEA (carciroma embryonic antigen) level assays respectively. Immunohistochemical localization of OEA in the tumor tissues were also determined. Eesults showed tha morphological and functional characteristics of the transplanted tumors to be basically identical to the parent tumor's cell.Radioimmunoimaging studies in the nude mice bearing transplanted tumora showed that satisfactory tumor imaging could be observed 72 hours after injection of 131I-CEA-McAb intraperitoneally. The tumor/intestinal ratio was 4.3. Labelled antibodies could accumulate in tumor tissue.All the experimental results showed that nude mice bearing the transplanted tumor can be used as an ideal experimental animal for related research of large bowel carcinoma.
