摘  要:目的 探讨中性鞘糖脂 (N GSLs)表达在卵巢癌细胞株的多药耐药 (MDR)中的作用。方法 利用三苯氧胺(TAM)和维拉帕米 (VRP)为逆转剂 ,运用MTT法分析了TAM、VRP的逆转效应。采用改良的Hakamori方法提取、纯化了耐药亚株SKOV3 /AdrMDR逆转前后及亲本株SKOV3 细胞的N GSLs ,高效薄层层析 (HPTLC)分析了这些N GSLs的表达。结果 SKOV3 /Adr和SKOV3 的N GSLs表达有差异 ,SKOV3 /Adr的CMH (monohexosylceramide ,单己糖神经鞘氨醇 )表达较SKOV3 强。经TAM、VRP作用后 ,SKOV3 /Adr的耐药性发生逆转 ,CMH表达降低。结论 卵巢癌的多药耐药性与中性鞘糖脂相关 ,CMH可能为卵巢癌MDR相关中性鞘糖脂抗原。Objective To evaluate the relationship between MDR of ovarian cancer and expression of neutral glycosphingolipids in ovarian cancer cell lines. Methods The effects of TAM and VRP(which have been shown to reverse MDR) on the growth of SKOV 3/Adr cell lines were assayed by MTT method. N-GSLs of the cells were isolated and purified by the modified Hakamoris method and analysed by HPTLC. Results The expression of N-GSLs was different between parent cell lines and resistant cell sublines,and the level of CMH was higher in SKOV 3/Adr than in SKOV 3. TAM and VRP could render multidrug-resistant cells sensitive to chemotherapy,while the level of CMH concomitantly had shown a sharp decrease. Conclusion The expression of N-GSLs is associated with MDR of ovarian cancer,and CMH might be a kind of MDR related glycolipids in ovarian cancer.
