摘  要:目的 通过研究人肾癌组织和癌旁正常组织中差异表达的基因 ,寻找肾癌相关基因以用于诊断和治疗。方法 以包含 80 0 0个cDNA基因表达谱芯片研究 1组肾癌组织样本的基因表达谱。按一步法抽提肾癌和对照组癌旁正常组织的总RNA并纯化mRNA ;将等量的对照组织和肾癌组织mRNA分别逆转录合成荧光分子掺入的cDNA—链做探针 ,混合后杂交上述基因芯片。经严格洗片后用ScanArray 3 0 0 0扫描仪扫描芯片荧光信号图像 ,计算分析后比较 2种组织中差异表达的基因。 结果 共筛选出差异表达基因 95条 ,其中未知基因 44条。结论 基因芯片在筛选肾癌相关基因的改变具有快速、高通量、灵敏的特点 ,肾癌在细胞代谢、信号转录。Objective To investigate the different genes expression in renal carcinoma and in normal tumour-surrounding tissue and to apply renal cell carcinoma associated genes in diagnosis and treatment.Methods Equal quantity mRNA from renal cell carcinoma and adjacent normal renal tissues were retro-transcribed and labeled with cy5 and cy3 fluorescence as probes.The mixed probe was hybridized with two piece H 80 gene chip.It was scanned by laser scanner.The acquired image was analysed.Results A total of 1333 genes were screened out whose expression level were more than 2 times in renal cell carcinoma as in adjactive normal renal tissue.44 new genes were found in the study.Conclusion The cDNA microarray for analysis of gene expression profile is a powerful method to identify renal cell carcinoma associated genes,which involved in cell cycle,signal transduction,protein synthesis and metablism.
