摘  要:目的 研究高强度聚焦超声 (HIFU )诱导人肺癌细胞凋亡的作用及可能机制。方法 用不同剂量 HIFU作用于人肺癌细胞株 A549,分别应用 MTT试验、集落形成试验及流式细胞仪检测细胞存活、增殖、凋亡及相关基因表达的改变。结果 一定剂量的 HIFU可通过影响细胞增殖和凋亡相关的基因 (p53、fas、bax及 HSP70 )表达 ,阻止细胞于 G0 / G1期 ,抑制细胞增殖和集落形成 ,诱导细胞凋亡。结论 一定剂量的 HIFU可抑制体外培养的人肺癌细胞增殖 ,诱导细胞凋亡 ,其作用机制可能与其调控细胞增殖。Objective To investigate high intensity focused ultrasound (HIFU)inducing apoptosis of hum an lung cancer cell line and to study the mechanism of its action. Metho ds Human lung cancer cells (A549) were treated with different intensit y of HIFU in vitro. The killing effect and proliferation inhibition of HIFU on c ancer cells were investigated with the methods of trypan blue exclusion assay, M TT assay and colony formation assay. And the cell cycle, apoptotic cells and the expressions of p53,bcl-2,bax,fas and HSP70 were measured with flow cytometry. Results After HIFU treatment, the death rates of cancer cells i ncreased, cell proliferation decreased(P<0 05), and the number of colony fo rmation decreased(P<0 001). The cell number in G 0/G 1 phase increased ( P<0 05) and in S phase decreased, the apoptotic index increased(P<0 01 ), the expression of p53, fas, bax, HSP70 gene increased(P<0 01), and the e xpression of bcl-2 gene had no significant difference(P>0 05). Co nclusion The results suggest that HIFU may inhibit human lung cancer ce ll proliferation and induce cell apoptosis. Its mechanism may be related to regu lating the genes of cell proliferation and apoptosis.
