摘 要:目的 探讨神经生长因子 (NGF)对角膜上皮细胞增殖的影响 ,为寻找促进角膜上皮损伤修复的药物提供依据。方法 在人和兔角膜上皮细胞培养液中加入NGF ,采用四甲基偶氮唑盐 (MTT)法观测细胞增殖情况。并用流式细胞仪检测NGF对人角膜上皮细胞增殖核抗原表达的影响。结果 加入外源性NGF后 ,培养的人和兔角膜上皮细胞增殖率较对照组均有明显增加 (P <0 0 1,P <0 0 5 ) ,流式细胞仪检测细胞增殖核抗原的表达较对照组明显增多(P <0 0 1)。Objective To study the effect of nerve growth factor (NGF) on proliferation of human and rabbit corneal epithelial cells. Methods The human and rabbit corneal epithelial cells were cultured, then treated with NGF. MTT assay was used to examine the proliferation of the cells. The effect of NGF on the expression of proliferating cell nuclear antigen in the human epithelial cells was observed by flow cytometry. Results After addition of exogenous NGF, the proliferative efficacy of the cultured human and rabbit corneal epithelial cells was markedly increased as compared with control group ( P <0 05) and the PCNA expression of the corneal epithelial cells cultured in NGF was also increased ( P <0 01). Conclusion Exogenous NGF can stimulate proliferation of human and rabbit corneal epithelial cells. NGF can serve as an effective preparation to repair the corneal injury.
