摘 要:目的 观察烹调油烟冷凝物 (CookingOilFumeCondensates,COFC)的体外发育毒性并探讨其可能的毒性作用机制。方法 采用体外全胚胎培养模型 ,将植入后 8 5d龄昆明种小鼠胚胎在大鼠即刻离心血清培养基中培养 48h(COFC染毒浓度分别为 0、2 0、40、80、160、3 2 0mg/L) ,观察COFC对小鼠胚胎生长发育和组织器官形态分化的影响。结果 COFC在≥ 80mg/L以上剂量时能抑制胚胎生长发育和形态分化 ,诱发卵黄囊生长和血管分化不良 ,发育异常率增高 ;高浓度组能诱发多种畸形 ,主要为神经系统 ,心脏、肢芽等器官系统畸形。结论 体外实验条件下 。Objective To observe the developmental toxicity and to explore the possible mechanisms of the cooking oil fume condensates (COFC) on mouse embryos in vitro . Methods The postimplantation whole embryo culture system was used. The day 8 5 isolated KM mouse embryos (2-5 somites ) at organogenetic stage were cultured for 48 hours in rat immediately centrifugal serum (ICS) with COFC ( final concentration 0,20, 40, 80, 160 and 320 mg/L ) in the presence of rat liver S 9 mix. Results COFC at high level (≥80mg/L) could inhibit the growth and blood vessel differentiation of visceral yolk sac (VYS) and increase abnormal embryos. The major abnormalities included abnormal nervous system (small forebrain and midbrain, open neural tube, encephalocele, hydrocephaly), abnormal arch and heart (retarded heart, pericardial edema), small optic, abnormal flexions, small forelimb bud and so on. Conclusion It was concluded that , to some extent, COFC had embryo toxicity and teratogenicity on cultured mouse embryos in vitro.
