短裙竹荪菌丝体多糖DdM-S提取及其性质

摘　　要：短裙竹荪菌丝体经热水提取 ,乙醇沉淀 ,去蛋白后得到粗多糖 ,经DEAE -SephadexA - 2 5柱层析进一步纯化 ,得到多糖纯品DdM -S。DdM -S经聚丙烯酰胺凝胶电泳和SephadexG - 2 0 0柱层析 ,表明该多糖为单一物质。纸层析和气相层析分析表明 ,DdM -S的单糖组成为D -葡萄糖 ,D -甘露糖和L -岩藻糖 ,其摩尔比为1 2 8∶1 0 0∶0 36。多糖的分子量约为 199KD ,红外光谱呈现出典型多糖的吸收峰。A polysaccharide DdM-S was obtained through being extracted from the submerged mycelium of Dictyophra duplicata by hot water,precipitated by ethanol,deproteined by protease method and Sevag method,and further puried by DEAE-Sephadex A-25 column.DdM-S was proved homogeneous by polyacrylamide gel electrophoresis and chromatography on Sephadex G-200.Identified by paper chromatography and gas chromatography,it was proved that the components of DdM-S were composed of D-Glc,D-Man and L-Fui with a molar ratio of 1.28∶1.00∶0.36 and that its molecular weight was estimated to be 199KD.Typical absorption of polysaccharide DdM-S was shown in its IR spectrum.
