摘 要:高产缬氨酸的北京棒杆菌(Corynebacterium pekinense)突变株125菌株,在2.6L自控发酵罐上分批培养结果表明,当发酵中后期DO为零时,产酸量较多。也可用k_(L2)值为指标来调节过程的供氧强度,k_(La)=90.75h^(-1)时,产酸量最高。同时比较了恒速连续补加葡萄糖液,当F=3.75g/h时,产酸较高。由此获得了该菌株L-缬氨酸发酵的低供氧与恒速补糖的控制模式。总糖量为16.85%的发酵,可使产酸量达38.2g/L,转化率为22.67%。本文对有关试验结果,进行了发酵动力学的分析和讨论。Study on L-valine fermentation were made in a 2.6 L jar fermentor using a valine producing mutant of Corynebacterium pekinese 125. The batch culture congirmed that the valine fermentation belonged to the kinetic type Ⅱ of fermentation and the oxygen supply greatly affected the production of valine. The influence of the dissolved oxygen tension on the valine fermentation was investigated. The valine formation was maximum when the DO approcbed zero twenty hours later after the fermentation start. By means of the control of k_La, the oxygen supply was controled. The maximun productivity occurred when h_La=90.78h. The batch culture also indicated that the higher glucose concentration in culture broth inhibited the ability of the mutant for valine formation and decreased the product yield. The possibility of improving valine production was then studied using fed-batch culture. Several constant feed rates were tested. Initial glucose concentration 60g/L, was chosed in the batch period of the designed fed-batch culture. When the glucose conc. was reduced to 10-20g/L, the additional glucose solution began to be added into the fermentor. The higher production was obtaned at the feed rate of 3.75 g/h (glucose solution conc. of 420g/L). Finally, a control strategy for the supply of oxygen and glucose in the fed batch culture valine fermentation was developed, i.e. the control method of low dissolved oxygen tension and the constant glucose feed. Using this strategy, the production of valine was imporved to 38.2g/L, and the product yield was 22.75% with total glucose concentration of 177.7g/L. And this strategy provided a base of the valine fermentation scal-up.
