摘 要:目的 :观察苯并 [a]芘诱发的细胞应答反应 ,探讨苯并 [a]芘引发的基因突变和致癌机制。方法 :应用双向凝胶电泳与相应软件分析苯并 [a]芘处理后细胞内蛋白质的表达谱 ,MAL DI- TOF(matrix assisted laser desorp-tion/ ionization- time of flight)质谱结合数据库检索鉴定差异表达的蛋白斑点。结果 :细胞经苯并 [a]芘处理后 ,可引起广泛的蛋白质表达的改变 ,其中以锌指蛋白和一些参与转录调控的蛋白多见。结论 :这些表达改变的锌指蛋白和转录调控因子参与了苯并 [a]芘诱发细胞的应答反应 ,提示可能参与苯并 [a]芘引发的基因突变和肿瘤形成。Objective: To understand benzopyrene (BP) mediated cellular responses,and to provide clues to explore molecular mechanism of mutagenesis and carcinogenesis induced by BP. Methods: Two-dimensional electrophoresis (2-DE) was used to investigate the protein expression levels of FL cells after BP exposure,matrix-assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF-MS) combined with database search was applied to identify the differentially expressed proteins. Results: Statistical analysis showed that the volumes of 47 protein spots were altered after BP treatment (P<0.05) and 23 proteins were successfully identified,including zinc finger proteins,SWI/SNF related protein,Bromo-domain containing domain and other proteins. Conclusion: These affected proteins may be involved in the cellular responses to BP exposure,and may mediate the BP induced mutagenesis and carcinogenesis.
