摘  要:目的 观察成纤维细胞热烫伤后丝裂原活化蛋白激酶以及活化蛋白 1的表达。方法 将培养的人成纤维细胞分成 4组 :( 1)单纯热损伤刺激组 ;( 2 )热损伤刺激 +碱性成纤维细胞生长因子处理组 ( 10 μg/L) ;( 3 )预先加入PD980 5 9( 10 μmol/L) ,再行热损伤 +碱性成纤维细胞生长因子 (bFGF)刺激 ;( 4 )预先加入PD980 5 9+SB2 0 3 5 80 (各 10 μmol/L)阻断剂 3 0min ,再行热损伤 +碱性成纤维细胞生长因子刺激。伤后 0、3 0、60和 180min检测细胞外信号调节激酶 (ERK )和原癌基因c fos蛋白。结果 单纯热刺激的成纤维细胞ERK表达增加 ,随后消失。碱性成纤维细胞生长因子刺激ERK表达增加显著 ,时间延长。加入PD980 5 9拮抗剂 ,ERK的表达受抑制。原癌基因c fos的表达开始增加 ,随后减弱。同时阻断ERK和 p3 8MAPK两条通路 ,c fos弱阳性表达。 结论 碱性成纤维细胞生长因子引起热损伤成纤维细胞ERK表达增加 ,原癌基因c fos是MAPK信号通路下游重要的靶蛋白。Objective To explore the change in mitogen-activated protein kinases (MAPKs) in scalded fibroblasts and the expression of activated protein-1.Methods Human dermal fibroblasts were divided into four groups:control group (A);Basic fibroblast growth factor (bFGF,10 μg/L)+heat stress (B);PD98059 (10 μmol/L)+bFGF+heat stress;D:PD98059+SB203580 (10 μmol/L each)+bFGF+heat stress.Westren blotting was performed to detect the expression of ERK1/2 and oncogene c-fos.Results The expression of ERK1/2 was increased in group A and then gradual disappeared.ERK expression was markedly stronger after being treated with exogenous bFGF.PD98059 could inhibit the expression of ERK.The expression of oncogene c-fos was increased,and became evidently weak at 180 min.However,the expression could last up to 180 min when bFGF was added.Inhibition of ERK activation could not depress c-fos expression.MAPK signal pathways were inhibited,and the c-fos expression became weak.Conclusion The expression of ERK was markedly increased in heat-stress fibroblasts after being stimulated by exogenous bFGF.c-fos played a key role as a down-stream protein in MAPK signal pathway.
