乳腺癌中端粒酶活性半定量及其与病理学指标的关系

摘　　要：目的　探讨乳腺癌中端粒酶 (TLMA)活性表达及其与病理学指标的关系。方法　运用银染端粒重复序列扩增法 (SS TRAP)检测了 5 2例乳腺癌组织中TLMA活性 ,并与癌旁、良性及正常乳腺组织进行对照 ,其结果予以半定量。结果　乳腺癌组TLMA阳性表达率为 90 .3 8% ,明显高于癌旁、良性及正常组 ( 2 8.85 %、3 1.2 5 %、0 ,P均 <0 .0 1)。Radit分析结果显示 ,乳腺癌中TL MA的表达与病理类型、临床分期、肿瘤大小、分化程度、淋巴结转移状况明显相关 (P <0 .0 5 ) ,而与患者病程、年龄因素无关 (P >0 .0 5 )。结论　半定量SS TRAP法是一种简便易行、敏感特异的TLMA活性检测方法 ;TLMA的激活可能发生在乳腺癌的早期阶段 ,并在其发生发展过程中起重要作用。Objective To investigate the expression of telomerase activity and its relationship with pathological indexes in breast carcinoma.Methods the telomerase activity was detected in 52 breast carcinoma specimens by silver staining telomeric repeat amplification protocol assay (SS-TRAP) and compared with that in paracarcinoma tissues,benign lesions and normal galactophore tissues,The results were analyzed by semi-quantification.Results The positive frequency of telomerase expression in breast carcinoma was 90.38 %,significantly higher than that in paracarcinoma (28.85 %),benign lesions (31.25%) and normal mammary tissues (0) (all P <0.01).The Radit analysis revealed that the expression of telomerase in breast carcinoma was obviously correlated with clinopathological categories,clinical grade,tumor size,degree of differentiation and status of lymph metastasis (all P <0.05),but had no correlation with courses of disease or age (all P >0.05).Conclusion Semi-quantificative assay based on SS-TRAP was feasible and simple,sensitive and special for detection of the expression of telomerase.The activation of telomerase might occur at the early stage of breast carcinoma and play an important role during the process of carcinogenesis and development.
