摘 要:目的 探讨人白细胞抗原(HLA)Ⅰ类和Ⅱ类分子在肝细胞癌(简称肝癌)的表达情况及其意义.方法 采用免疫组化法分别检测HLA-ABC和HLA-DR抗原在46例人肝癌组织、10例人正常肝组织以及SMMC-7721、HHCC、HCC-9204和BEL-7402等4种人肝癌细胞系和人正常肝细胞系QZG的表达情况,并以图像分析仪进行定量分析.结果 肝癌和正常肝组织及其相应细胞系的HLA-ABC和HLA-DR抗原阳性信号均主要定位于细胞浆.肝癌组织表达HLA-ABC的强度明显弱于正常肝组织(P<0.05),而对HLA-DR的表达明显强于正常肝组织(P<0.05).4种肝癌细胞系的HLA-ABC表达强度均明显低于QZG细胞(P<0.05),但对HLA-DR的表达与QZG细胞末见明显差异(P>0.05).结论 肝癌组织和肝癌细胞系中HLA-ABC抗原表达有所下降.肝癌组织有HLA-DR抗原表达增强,而肝癌细胞系基本上不表达HA-DR.To require into the expression of HLA-Ⅰ and HLA-Ⅱ molecules in hepatocellular carcinoma (HOC) and its significance. Methods: The expression of HLA-ABC and HLA-DR antigens in 46 human HOC tissues, 10 human normal liver tissues, 4 human HOC cell lines SMMC-7721, HHCC, HCC-9204 and BEL-7402, and a human normal liver cell line QZG was detected with immunohistochemistry, and analyzed with image-quantitative analysis. Results: Most of the positive signals of HOC and normal liver tissues and their cell lines located within cytoplasm. The intensity of HLA-ABC expression In HOC tissues was weaker than that in normal liver tissues significantly (P<0.05), but the intensity of HLA-DR expression was stronger than that in normal liver tissues significantly (P< 0.05). The intensity of HLA-ABC expression in 4 HOC cell lines was weaker than that innormal liver cell line significantly (P<01.05),but there wasn't significant difference on the intensity of HLA-DR expression between HCC and normal liver cell lines (P>0.05). Conclusion: There is decline of HLA-ABC antigen expression in HCC tissues and HCC cell lines. There is ennancement of HLA-DR antigen in HCC tissues, but HOC cell line can't express HLA-DR on the whole.
