改良MTT法原代癌细胞培养药物敏感性试验的实验研究

摘　　要：目的　研究改良MTT法原代癌细胞药物敏感性试验及影响因素。方法　采用淋巴细胞分离液梯度离心法分离纯化癌细胞 ,MTT比色法测定 4 5例 (胃癌 15例、肺癌 10例、大肠癌 12例、乳腺癌 8例 )体外原代癌细胞对 8种常用化疗药物的敏感性。结果　采用 3 0分钟消化时间、10 0 %～ 60 %密度梯度淋巴细胞分离液以及标本离体后即刻实验 ,活细胞百分率为 94 65± 2 8 4 7,药敏阳性率为 71 11% ,与其它实验条件相比差异显著 (P <0 0 1)。结论　改良MTT法是一种简便、快捷的肿瘤体外药敏试验方法 ,为临床实施个体化化疗提供了实验依据 ,值得在临床工作中推广应用。Objective Study the changed method of the anticancer drug's sensitivity in the cancer cells with MTT colormetric assay and it's effictive factor. Methods Using density gradient centrifugation for seperating cancer cell.45 prime cancer cells of patients in vitro (15 of samples gastrics cancer, 10 samples of lung cancer, 12 samples of colon cancer, 8 samples of breast cancer) were examined with MTT assay to find out their drugs' sensitivity to the often used chemotherapy medicine Results Using 30 minutes digestion time, 100%～60% concentration of lymphcyte seperationg liquid and doing the assay as the sample obtained as possible, the percentage of living cancer cells was 94 65 ±28 47,positive druges sensitivity which is 71 11% is significant different with other group. Conclusion The changed method of MTT assay is a easy and quick method for drugs' sensitivity test in vitro and it provides experimental proof for individual chemotherapy This changed method is deserved in using in clinical routine.
