摘  要:目的 探讨胃癌及其癌前病变线粒体DNA微卫星不稳定 (mtMSI)与Bcl 2、BaxmRNA表达的关系。方法 采用PCR SS CP方法检测mtMSI,并采用RT PCR方法检测了Bcl 2和BaxmRNA的表达。结果 胃黏膜肠化 (5 3 3% )、异型增生 (70 % )组织Bcl 2mRNA表达率显著高于浅表性胃炎组(10 % ) (P <0 0 5 ) ,而慢性萎缩性胃炎 (5 0 % )和胃癌 (30 % )Bcl 2mRNA表达率与浅表胃炎组相比差异无统计学意义 (P >0 0 5 ) ;胃黏膜异型增生组织Bcl 2mRNA表达率显著高于胃癌组织 (P <0 0 5 )。异型增生 (6 0 % )组BaxmRNA表达率显著高于浅表胃炎组 (P <0 0 5 ) ,而慢性萎缩性胃炎(5 0 % )、肠化 (4 6 7% )和胃癌 (33 3% )的BaxmRNA表达率与浅表性胃炎组 (10 % )比较差异无统计学意义 (P >0 0 5 )。mtMSI(+)组Bcl 2mRNA表达率为 5 0 % ,mtMSI(- )组为 4 0 5 % ;mtMSI(+)组BaxmRNA表达率为 4 4 4 % ,mtMSI(- )组为 4 0 5 % ,mtMSI(+)组Bcl 2和BaxmRNA表达率与mtMSI(- )组相比差异无统计学意义 (P >0 0 5 )。结论 mtMSI可能在部分胃癌的发生中起重要作用 ;胃癌发生过程中出现的mtMSI与Bcl 2。Objective To explore the relationship between mitochondrial DNA microsatellite instability(mtMSI) and expression of Bcl 2 and Bax mRNA in gastric cancer and its precancerous lesions. Methods mtMSI and expressions of Bcl 2 and Bax mRNA were detected with PCR SSCP and RT PCR. Results Expressions of Bcl 2 mRNA in intestinal metaplasia (IM,53 3%) and dysplasia (Dys, 70%) were significantly higher than that in normal control(10%, P <0 05), whereas no significant differences were found among chronic gastritis (CAG, 50%), GC (30%) and normal control ( P <0 05) Expression of Bcl 2 mRNA in Dys was higher than that in GC( P <0 05). Expression of Bax mRNA was significantly increased in Dys (60%) ( P <0 05), but not in CAG (50%), IM (46 7%) and GC (33 3%)( P <0 05), compared with normal control (10%). Expression of Bcl 2 and Bax mRNA in H.pylori infected gastric mucosa was significantly higher than that in non H.pylori infected gastric mucosa( P <0 05), whereas expressions of Bcl 2 and Bax mRNA were not consistent with H.pylori CagA status ( P <0 05). No significant difference was found between expression of Bcl 2 and Bax mRNA in mtMSI(+) and that in mtMSI(―) ( P <0 05). Conclusion mtMSI may play an important role in some gastric cancers, and increased mtMSI is independent of abnormal expression of Bcl 2 and Bax mRNA.
