狂犬病病毒ERA株在犊牛睾丸细胞(BTC)上连续传代的稳定性及毒种批的制备与检验

摘　　要：将狂犬病病毒ERA株接种犊牛睾丸细胞进行了 2轮传代 ,并逐代测定毒价 (LD50 )。结果 :第一轮从 1 5代传至 2 6代 ,其毒价均 >1 0 4 .0 LD50 /0 .0 3mL ,但 2 0代以后似有下降趋势 ;第二轮从 1代传至 2 8代 ,9～ 2 2代毒价为 1 0 4 .5～ 1 0 5 .33LD50 /0 .0 3mL ,2 3代为 1 0 4 .0 LD50 /0 .0 3mL ,2 7、2 8代均 <1 0 4 .0 LD50 /0 .0 3mL。将第 6、1 6、2 8、2 9代ERA/BTC培养物和第 3代ERA/BHK 2 1细胞培养物分别作电镜负染观察 ,比较病毒颗粒形态 ,未见明显差异。将第 1 1代ERA/BTC培养物经冷冻真空干燥制成毒种批 ,其病毒含量为 1 0 5 .2 2 LD50 /0 .0 3mL ,水分≤ 2 .38% ,安全性、免疫原性。The Rabies virus (ERA strain) was inoculated on BTC cells. After 2 series of passages, the titers of the cultures were detected with LD 50 . In the first series from 15th to 26th passages, the titers of the cultures were more than 10 4.0 LD 50 /0.03 mL, while that of the 20th passage were reduced. In the second series from the first to 28th passages, the titers of the cultures was 10 4.5 ～10 5.33 LD 50 /0.03 mL from 9th to 22th passages, but the titers of the 23rd passage cultures were 10 4.0 LD 50 /0.03 mL and less than 10 4.0 LD 50 /0.03 mL for the later passages. After several passages on the BTC cells, the ERA/BTC cultures of the 6th, 16th, 28th, 29th passages and the ERA/BHK 21 culture of the third passage were inspected with electron microscope. There were no virus morphologic differences among them. The 11th passage of the ERA/BTC cultures was freezedried as the seed lots and detected. The virus titers were 10 5.22 LD 50 /0.03 mL, the residual moisture was not more than 2.38%. The safety, immunogenicity, specificity and purity of the seed lots were all satisfied.
